It could be something simple, that is easy to overlook. Are the players in adventure mode? (if they are, I suggest you look in server properties and change the gamemode [don't worry if this is the case, it is an easy mistake to make]) If not, it could be related to worldguard. I am assuming you selected the region, and so if it is related to worldguard, it will probably be a permission. If it is not either of those, it will probably be something big, scary and complicated :/
