It just depends on what the thing is really like lets say its a promotion for rank donor most permissions plugins I have seen require the player to be online But if you mean just how to set that in general I believe there is a checkbox for if it is required that they are online or not
I seem to be able to add players to a group while they're offline. I'm using group manager. For buycraft my initial command would be /manuadd <player> <group>
